Module Catalogues, Xi'an Jiaotong-Liverpool University
Module Code: BIO205
Module Title: Molecular and Cell Biology Techniques
Module Level: Level 2
Module Credits: 5.00
Academic Year: 2020/21
Semester: SEM1
Originating Department: Biological Sciences
Pre-requisites: N/A
This module aims at:

- Providing practical experience in many of the techniques currently used in molecular biology. This will include introduction to the skills and techniques necessary to isolate, clone and analyse genes.

- Providing practical and theoretical experience in techniques currently used in cell biology.
Learning outcomes
At the end of this module students will acquire knowledge and critical understanding of concepts and principles of the following techniques:

A : -Analysis of DNA fragments by agarose gel electrophoresis and restriction mapping.

B: -PCR based assays for gene cloning or microsatellite analysis.

C: -Methods used for cloning, sequencing and database analysis of genes.

D: -The effects of external factors such as temperature and pH on the growth and physiology of biological systems.

E: The culture conditions required by different biological systems.

F: -The application of computers in modern biology.

G: They will also acquire ability to: -Present, critically evaluate and interpret qualitative and quantitative data, and record procedures and protocols. -Make sound judgements based on basic theories and concepts. -Develop team-working and individual skills and also learn to manage time effectively. -Plan and execute a series of experiments, use laboratory equipment correctly and safely to generate data.

Method of teaching and learning
This practical course will be conducted in Biological Sciences laboratories. Each session will be preceded by a lecture session to provide background in theory and procedures, and followed by a lecture/workshop session to discuss learning outcomes. In the laboratory session students will work individually or within small groups and receive help in the planning, execution and interpretation of the experiments from demonstrators. Course manuals will provide theoretical background, detailed protocols and procedures, and examples of worked data.
Molecular biology techniques: Assessment of skill level (automatic pipettes, molarity calculations). Bacteriological techniques, transformation and plasmid DNA isolation. -Plasmid DNA analysis by restriction enzyme digestion and agarose gel electrophoresis. -Gene cloning. -Determining the size of plasmid inserts using the Polymerase Chain Reaction (Colony PCR). -Data handling session testing previously taught analysis skills. Microbiological techniques: -Response of E. coli to stress. Design and execution of neutrophil model and acid stress experiments. -Behavioural responses of Dictyostelium discoideum. Slime mould development, phototaxis and phagocytosis; scientific illustration. -Fixation of marine phytoplankton: methods and applications; image analysis. Analysis of data and report writing. -Identification of nephrons in mouse kidney. H & E stain; immunochemistry; lectin staining
